According to the CDC Fact Sheet for Sheltering In Place during Radiation Emergency;

§“When you move to your shelter, use duct tape and plastic sheeting to seal any doors, windows, or vents for a short period of time in case a radiation plume is passing over (listen to your radio for instructions). Within a few hours, you should remove the plastic and duct tape and ventilate the room. Suffocation could occur if you keep the shelter tightly sealed for more than a few hours. “

Shortcomings of Duct Tape, Plastic

The Office of Homeland Security recommended that citizens prepare for possible nuclear, biological or chemical attack by, among other things, having a supply of duct tape and plastic sheeting that could be used to seal up a room, making it more resistant to such agents.

While this approach might be effective to a point, there are shortcomings.

   It is very difficult to create a perfectly sealed room using these materials, especially if the work is done hurriedly.
   Normal plastic sheeting may not be resistant to certain gasses.
   Even if a room could be thoroughly sealed, the air supply inside would be limited.

The problem is not so much running out of oxygen, but rather the build-up of carbon dioxide and water vapor. At best, a well-sealed room might provide protection for a few hours before it became intolerable, especially for children and older individuals.
